Actim®Calprotectinisarapidimmunologicaldipsticktestthatisbasedonhighlyspecificmonoclonalantibodiesdetectingfecalcalprotectin.Thetestkitcontainsallnecessarymaterialsforhomesampling,andtheone-steptestprotocolrequiresonly15secondsofhands-ontime.Theresultscanbereadfromtheteststripinjust10minutes:thenumberofbluelinesindicatestheconcentrationrangeofcalprotectininfeces.
ActimCalprotectingivessemiquantitativeresultsbyreportingcalprotectinconcentrationinthreeranges:<50,50–200,and>200μg/g.Thishelpstodistinguishbetweenpatientswithoutmucosaldamage,thosewithanirritablebowel,andthosewithactiveinflammation.

ABOUTCALPROTECTIN
- FecalcalprotectinisasensitiveMarkerofintestinalinflammation.
- Itisanimmunomodulatoryandantimicrobialproteinthatisprimarilysecretedbyneutrophils.Mucosaldamagecausesneutrophildegranulation,whichleadstoelevatedcalprotectinconcentrationsinthebowelandfeces.
- Calprotectinisanextremelystableprotein,andcanbefoundinfecesevenafter7daysofsecretionfromthemucosalmembrane.

FecalcalprotectinisanimportantbiomarkerforIBD,includingCrohn’sdiseaseandulcerativecolitis.Calprotectinlevelsarelowinfecesofhealthyindividuals,whileIBDpatientshaveelevatedfecalcalprotectinconcentrationsduetoleakagefromdamagedmucosa.TheActim®CalprotectintestisausefultoolfordistinguishingIBDfromIBS,whichdoesnotinvolvepathologicaldamagetothebowel.

HowtouseActimCalprotectin
ActimCalprotectintestkitcontainsallnecessarymaterials,andcanbestoredatroomtemperature(2–30°C).
- Collectsample
OpentheSpecimenDilutionBuffertubeandtakesomefecesusingthesamplingstickattachedtothescrewcap.Twistthestickindifferentplacesinthefecessample.
Putthesamplingstickbackintothetubebypushingthesticktroughtheconeandclosethecaptightly.ShakethetubetosUSPendthespecimeninthebuffer.
- Perforation
Usethesharptipoftheperforatingdevice(dipstick)topuncturetheperforationareainthebottomofthebuffertubethenpushthedeviceintothetubeuntilthestopperpreventsitgoingfurther.
- Activatethetest
Slowlyinvertthetubeonceandimmediatelyplacethetubeonthetableinanuprightposition.
Itisimportantthattheinvertingmovementisnottooabrupt,apeacefulinversionisvitalforthecorrectperformanceofthetest.Also,thetubemustnotremainupsidedown.
- Interpretresults
Atestresultcanbereadin10minutesaftertestactivation.
